Boehringer bets up to $1B on RNA splicing targets from Envisagenics

The New York company will mine alternative splicing for tumor-specific targets that drug makers can turn into therapies.

For the next several years, Boehringer Ingelheim will search for cancer targets in the way cells cut and rejoin their RNA. Its partner is the New York company Envisagenics, whose AI platform, SpliceCore, points at alternative splicing, a process that lets one gene yield several distinct proteins. Terms were not disclosed, but Envisagenics said the collaboration could bring it more than $1B in upfront payments, research funding, option fees, milestones and royalties.

Targets that appear on cancer cells and nowhere else are rare in solid tumors. That scarcity is why several aggressive cancers still have few good treatment options.

Boehringer can pursue the validated targets through several formats, including antibody-drug conjugates, T-cell engagers and multispecific antibodies. That range matters because it lets one target feed more than one program.

Splicing has drawn interest because it multiplies the number of possible targets without adding genes. A single transcript can yield a cancer-specific variant that healthy tissue rarely presents, which gives developers a narrower window for toxicity.

Why it matters. Target discovery, not molecule design, is the bottleneck that limits most AI drug platforms. Envisagenics sells a way to manufacture targets. If Boehringer’s teams convert even a few into clinical candidates, the company becomes a supplier to the wider industry rather than a single pipeline.
